PELE OLEOSA E HIPERSEBORREIA

A pele oleosa é caracterizada pelo acúmulo na sua superfície do excesso de sebo, uma substância secretada pelas glândulas sebáceas. O sebo é secretado pelos sebócitos por meio de um processo de diferenciação celular e da síntese e acúmulo de lipídeos, chamados de lipidogênese.

A lipidogênese é dependente de muitos fatores (hormônios, alimentação, ingredientes ativos farmacêuticos, xenobióticos etc.), que podem ser identificados em experimentos in vitro. Os sebócitos maduros são submetidos à apoptose e ruptura para liberação do sebo, que viaja do folículo piloso até a superfície da pele. O sebo lubrifica o folículo piloso e se espalha na superfície da pele no nível do estrato córneo.

A superprodução do sebo, chamada hiperseborreia, é responsável pela pele oleosa e pelo cabelo oleoso. Em alguns casos, a hiperseborreia também pode contribuir para o desenvolvimento da pele com tendência à acne.

Pele oleosa e hiperseborreia: modelos e ensaios in vitro

We analysed the activity of reference anti-androgens (Finasteride, Dutasteride, Cyproterone acetate), at the level of their potential targets (5-alpha-reductase, AR translocation, induced transcripts) and in a functional lipid accumulation assay. We also evaluated this androgen-induced lipid accumulation assay as a potential pharmacological tool to detect potential inhibitors from both androgenic and non-androgenic origins.

In order to study the impact of active androgens in sebocytes, we constructed a stable human sebocyte cell line derived from SEBO662 [17] constitutively expressing a fully functional AR. In these SEBO662 AR+ cells, dihydrotestosterone (DHT) induced AR nuclear translocation and the strong modulation of a set of transcripts (RASD1, GREB1…) known to be androgen-sensitive in other androgenic cells and tissues.
